If they load the ads via JavaScript, the js can detect if the load was successful.

So if I block that script at the DNS level, the site will report that the script failed to load. This tips of the ad was blocked. Am I correct?

Yes. For what it's worth, I've personally not seen it done in a way that is integrated with long-lived tracking mechanisms, that would explicitly flag you as a "DNS-ad-blocking-person," but I wouldn't put it past Google and others to do it.
