There is no sure-fire way to know, but there are some indications that may point to cheating. I'm going to leave each one with an alternate explanation or three.
Sudden decrease in sexual interest in partner (could also result from new medications or dosages, as well as other life changes)
Sudden increase in sexual interest in partner (could also result from new medications or dosages, as well as other life changes)
Unfounded accusations of cheating (could also be a sign of lost of trust, etc.)
A new tendency toward secretiveness (could also be a sign of addiction, alienation, etc.)
A new, unexplained tendency to spend time away from home (could also be a sign of addiction, alienation, etc.)
In short, look out for unexplained, major changes in behavior that could be signs of cheating (suddenly wanting to eat crescent rolls for every meal--probably not a sign of cheating, unless it's with the Pillsbury Doughboy). Also, please have some evidence before making accusations. Accusations of cheating can have a damaging effect on a relationship, and there's no need to inflict damage without cause.
