When I was an active duty Marine, it meant, one in the chamber, safety engaged. The saying may have originated when guns had external locks, but I don't really know.

A lock is the bit that holds a flint or match in a flintlock or matchlock firearm. Now we use percussive primers (usually) detonated by action of a hammer dropping with the force applied by means of a mainspring.
